Police were called to the two-vehicle crash on State Highway 63 west of Blenheim just after 6pm.
They said one person died at the scene and another had died from their injuries a short time ago.
A third person has been flown to Wellington Hospital with serious injuries.
Police said the road remained closed and motorists were asked to avoid the area as there was no diversion.
